Experimental Guidance and Technical Tips
- The immunogen was a recombinant fusion protein covering the full-length human SIRT6 (amino acids 1–355). This design may confer high specificity for native SIRT6 across assays.
- In Western blot, the predicted molecular weight is ~39 kDa (UniProt), with observed bands typically around 36–39 kDa. Validate detection in your experimental system and consider using appropriate loading controls.
- For immunoprecipitation (IP) and immunofluorescence (IF/ICC), we recommend titrating the antibody concentration and including proper positive/negative controls relevant to SIRT6 expression.
- For immunohistochemistry on paraffin sections (IHC-P), antigen retrieval optimization may be necessary. Test both heat-induced and enzymatic retrieval methods.
- Cross-reactivity with mouse and rat SIRT6 allows seamless translational studies, but confirm reactivity in your specific tissues or cell lines.
