Biological Background: SIRT5 Function and Localization

  • SIRT5 (SIR2L5) is an NAD-dependent protein deacylase that primarily acts as a demalonylase, desuccinylase, and deglutarylase to remove malonyl, succinyl, and glutaryl groups from target proteins. Related references: PMID:21908771 PMID:22076378 PMID:24703693

  • It activates carbamoyl phosphate synthetase 1 (CPS1) via desuccinylation and deglutarylation, thereby contributing to the regulation of blood ammonia levels during prolonged fasting. Related references: PMID:22076378 PMID:24703693

  • SIRT5 desuccinylates and activates superoxide dismutase 1 (SOD1), leading to a reduction in reactive oxygen species. Related references: PMID:24140062

  • It mediates desuccinylation of serine hydroxymethyltransferase 2 (SHMT2), thereby activating this enzyme. Related references: PMID:29180469

  • SIRT5 modulates ketogenesis through desuccinylation and activation of 3-hydroxy-3-methylglutaryl-CoA synthase 2 (HMGCS2) (By similarity).

  • The protein localizes predominantly to the mitochondrial matrix, but has also been detected in the mitochondrial intermembrane space, cytosol, and nucleus.

  • UniProt keywords for SIRT5 include Alternative splicing, Cytoplasm, Metal-binding, Mitochondrion, NAD, Nucleus, Proteomics identification, Reference proteome, Transferase, Transit peptide, and Zinc.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 37-310 of human SIRT5, covering the mature protein after cleavage of the mitochondrial transit peptide. This design should allow detection of both precursor and mature forms.
  • For Western blot, the predicted molecular weight is approximately 34 kDa. A band shift may occur upon mitochondrial import due to cleavage of the N-terminal transit peptide; validate in the relevant sample system.
  • The antibody is reported as KO validated, suggesting high specificity. Consider confirming with appropriate knockout cell lines or siRNA experiments for your model system.
  • For ELISA, use as a capture or detection antibody with recombinant SIRT5 protein or tissue lysates; optimize coating and detection conditions empirically.
