Biological Background: SIRT4 Function and Localization
- SIRT4, also known as SIR2L4, is a mitochondrial NAD-dependent protein lipoamidase, biotinylase, deacetylase, and ADP-ribosyltransferase.
- SIRT4 catalyzes the removal of lipoyl and biotinyl groups more efficiently than acetyl groups, inhibiting pyruvate dehydrogenase complex (PDH) through hydrolysis of lipoamide from the E2 component DLAT.

PMID:24052263 PMID:25525879 - It mono-ADP-ribosylates mitochondrial glutamate dehydrogenase 1 (GLUD1), inhibiting its activity and negatively regulating glutamine metabolism in response to DNA damage, leading to cell cycle arrest.
Related references:
PMID:16959573 PMID:17715127 - SIRT4 acts as a tumor suppressor; mTORC1 signaling represses SIRT4 expression, promoting anaplerosis and cell proliferation.
Related references:
PMID:23562301 PMID:23663782 - It mediates deacetylation of MLYCD at Lys-471, inhibiting its activity and thereby regulating lipid homeostasis.
- SIRT4 controls fatty acid oxidation by inhibiting PPARA transcriptional activation, likely through modulating NAD+ levels and disrupting SIRT1-PPARA interaction.
Related references:
PMID:24043310 - This protein is widely expressed, with prominent levels in vascular smooth muscle, striated muscle, and insulin-producing pancreatic beta-cells.
- It localizes to the mitochondrial matrix and is a zinc-dependent, NAD-dependent enzyme.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1-200) of human SIRT4; consider this epitope specificity when designing detection systems.
- For IF/ICC, optimize mitochondrial staining using appropriate fixation (e.g., 4% PFA) and permeabilization methods; validate with human-derived cell lines.
- In ELISA, this polyclonal antibody can be used as a capture or detection reagent; pair with a specific monoclonal antibody for sandwich assays if needed.
- As a polyclonal antibody, it may recognize multiple epitopes, offering robust signal but requiring careful validation for target specificity in complex samples.
- Confirm reactivity solely with human SIRT4; cross-reactivity with other species has not been reported.
