Biological Background: SIRT3 Function and Localization

  • SIRT3 (SIR2L3) is a mitochondrial NAD-dependent protein deacetylase and a member of the sirtuin family, featuring a conserved catalytic domain and a zinc-binding motif.
  • It deacetylates key lysine residues on numerous target proteins, including ACSS1, IDH, GDH, SOD2, PDHA1, LCAD, SDHA, MRPL12, and ATP5PO, thereby regulating energy metabolism. Related references: PMID:12186850 PMID:12374852 PMID:16788062 PMID:18680753 PMID:19535340
  • In response to metabolic stress, SIRT3 deacetylates FOXO3 and recruits FOXO3 and POLRMT to mtDNA to promote mtDNA transcription. Related references: PMID:23283301
  • SIRT3 regulates ceramide metabolism by deacetylating ceramide synthases CERS1, CERS2, and CERS6, increasing their activity and promoting ceramide accumulation.
  • It contributes to hepatic lipogenesis regulation by utilizing NAD+ imported via SLC25A47, triggering AMPK signaling and downregulating SREBP transcriptional activities to restore energy balance.
  • Beyond deacetylation, SIRT3 acts as a protein delactylase and debenzoylase, catalyzing delactylation of CCNE2 and histone H4K16, and debenzoylation of PPIF and ACLY. Related references: PMID:36896611 PMID:37720100 PMID:39524354
  • SIRT3 is localized to the mitochondrial matrix and is widely expressed across tissues.

Experimental Guidance and Technical Tips

  • The immunogen used was a synthetic peptide corresponding to amino acids 388-399 of human SIRT3 (NP_036371.1), located in the C-terminal region. When designing experiments, confirm that the epitope is accessible in your assay format.
  • For Western blotting (WB), the predicted molecular weight is approximately 44 kDa. Mitochondrial-rich fractions or whole-cell lysates from human, mouse, or rat cells can be used; optimize loading and transfer conditions.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), consider antigen retrieval methods and validate staining specificity using appropriate controls.
  • The antibody is suitable for immunoprecipitation (IP) and ELISA; for quantitative studies, titrate the antibody concentration and include relevant positive and negative controls.
