Biological Background: SIRT2 Function and Localization
- SIRT2 encodes a NAD-dependent protein deacetylase (sirtuin-2) that also functions as a protein-lysine deacylase, removing acetyl, benzoyl, methacryl, myristoyl, and palmitoyl groups from target proteins, including histones, α-tubulin, transcription factors, and metabolic enzymes. - Plays a pivotal role in cell cycle progression and genomic stability by functioning in the antephase checkpoint, preventing premature mitotic entry under microtubule stress, and positively regulating the anaphase promoting complex/cyclosome (APC/C) via deacetylation of CDC20 and FZR1. Related references: PMID:12697818, PMID:16909107, PMID:17488717, PMID:22014574
- Modulates chromatin structure and transcription by specifically deacetylating histone H4 at Lys-16 (H4K16ac) and histone H3 at Lys-56 (H3K56ac) during mitosis, facilitating H4K20 methylation; also associates with transcription start sites and enhancers of active genes. Related references: PMID:23468428, PMID:20587414
- Localizes to multiple subcellular compartments including the nucleus, cytoplasm, centrosome, spindle, midbody, and cell projections; its dynamic shuttling is critical for coordinating cell cycle-dependent functions and stress responses.
- Widely expressed, with isoform 1 prominent in heart, liver, and skeletal muscle, and isoform 2 enriched in the cerebral cortex; down-regulated in many gliomas and several other cancers, consistent with a tumor suppressor role.
- Participates in autophagy regulation by deacetylating FOXO1, and in metabolic control by stabilizing phosphoenolpyruvate carboxykinase PCK1 and activating glucose-6-phosphate dehydrogenase G6PD to manage glucose homeostasis and oxidative stress. Related references: PMID:20543840, PMID:21726808, PMID:24769394
- Deacetylates α-tubulin at Lys-40, thereby regulating microtubule dynamics, neuronal motility, oligodendroglial arborization, and neurite outgrowth; this activity is inhibited by phosphorylation at Ser-368 by the cyclin E-CDK2 complex. Related references: PMID:18332217, PMID:18995842, PMID:17488717
- Involved in innate immunity: upon Listeria monocytogenes infection, SIRT2 deacetylates histone H3 at Lys-18, dampening transcriptional activity to promote late infection stages. Related references: PMID:23908241
Experimental Guidance and Technical Tips
- The immunogen targets amino acids 300–350 near the C-terminus of human SIRT2; consider that isoforms or splice variants may differ in this region. Validate reactivity in your specific sample system.
- For Western blot (WB), expect a major band at approximately 43 kDa; use appropriate positive and negative controls. Optimize lysis and loading conditions to clearly resolve SIRT2.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), the antibody may detect nuclear and cytoplasmic staining depending on cell type and conditions; include antigen retrieval steps and confirm localization with known markers.
- This polyclonal antibody recognizes human, mouse, and rat SIRT2. Cross-reactivity with other species should be experimentally tested if required.
- For ELISA applications, consider pairing with a suitable capture/detection system; preliminary titration is recommended to establish optimal working concentrations.
