Biological Background: SIRT2 Function and Localization
- SIRT2 is an NAD-dependent protein deacetylase that removes acetyl groups from internal lysine residues of histones, alpha-tubulin, and numerous transcription factors, thereby modulating their functions. Related references: PMID:12620231, PMID:16648462, PMID:18249187
- SIRT2 plays a key role in cell cycle progression and genomic stability. It functions in the antephase checkpoint to prevent premature mitotic entry, and it positively regulates the APC/C complex by deacetylating CDC20 and FZR1, promoting mitosis. Related references: PMID:12697818, PMID:16909107, PMID:17488717, PMID:22014574
- SIRT2 deacetylates histone H4 at Lys-16 (H4K16ac) between the G2/M transition and metaphase, enabling subsequent H4K20 methylation, and also deacetylates histone H3 at Lys-56 (H3K56ac) during the G2/M transition, thereby regulating chromatin compaction and epigenetic signaling. Related references: PMID:23468428, PMID:20587414
- SIRT2 is involved in metabolic regulation: it deacetylates and stabilizes PCK1 to control blood glucose, activates G6PD in the pentose phosphate pathway to produce NADPH and counteract oxidative stress, and inhibits adipogenesis while promoting lipolysis. Related references: PMID:21726808, PMID:24769394, PMID:20543840
- SIRT2 also functions as a protein lysine deacylase, removing benzoyl, methacryl, myristoyl, and palmitoyl groups from target proteins like ARF6 and KRAS, regulating their membrane localization. Related references: PMID:30154464, PMID:34961760, PMID:25704306, PMID:29239724, PMID:32103017
- SIRT2 localizes to the nucleus, cytoplasm, perinuclear region, cytoskeleton (centrosome, spindle, midbody), and cell projections, consistent with its roles in mitosis, cell motility, and myelination. Two major isoforms exist: Isoform 1 is expressed in heart, liver, and skeletal muscle; Isoform 2 is strongly expressed in brain cortex. SIRT2 is frequently down-regulated in gliomas and certain cancers, and is considered a tumor suppressor.
Experimental Guidance and Technical Tips
- For western blotting (WB), a band near 43 kDa is expected for the canonical SIRT2 isoform. Alternative splicing and post‑translational modifications may yield additional bands; validate in your sample system.
- For immunohistochemistry (IHC-P), consider the broad subcellular distribution of SIRT2 and optimize antigen retrieval protocols accordingly. Tissues with high SIRT2 expression (e.g., liver, brain) can serve as positive controls.
- Given SIRT2’s cell cycle-dependent functions, synchronization of cell cultures may enhance detection in proliferation studies.
- For ELISA, ensure the capture/detection antibody format is compatible with the peptide‑immunized monoclonal antibody; note that the immunogen lies in the C‑terminal region of the protein.
- This antibody detects human, mouse, and rat SIRT2; cross‑reactivity with other species or sirtuin family members should be evaluated empirically.
