Use appropriate loading controls and validate band specificity with siRNA knockdown or knockout lysates if available.
  • SIRT1 is widely expressed but at low levels in liver and skeletal muscle; consider using tissue extracts known to express SIRT1 (e.g., brain, testis) for initial validation.
  • For ELISA applications, the peptide immunogen may provide good direct or capture detection sensitivity; optimize coating conditions and antibody dilutions.
