Biological Background: SIPA1 Function and Localization
- SIPA1 (Signal-induced proliferation-associated protein 1), also known as GTPase-activating protein Spa-1 or p130 SPA-1, is a 112 kDa protein encoded by the SIPA1 gene (Gene ID: 6494).
- Functions as a GTPase activator for Rap1 and Rap2, converting them to the inactive GDP-bound state.
Related references: PMID:9346962 - Involved in regulating cell cycle progression (by similarity).
- Localizes to the nucleus, cytoplasm, perinuclear region, and endomembrane system.
- Highly expressed in lymphoid tissues (thymus, spleen, peripheral blood lymphocytes) and also significantly in the spinal cord.
- Contains a coiled-coil domain, is subject to phosphorylation, and belongs to the human reference proteome.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 763–1042 of human SIPA1, targeting the C-terminal region; therefore, the antibody may detect full-length SIPA1 as well as any C-terminal fragments or isoforms containing this sequence.
- For western blot (WB), expect a major band at approximately 112 kDa. Optimize sample preparation and electrophoresis conditions to resolve this high-molecular-weight protein.
- Confirm species cross-reactivity in the relevant cellular context; the antibody is reported to react with human and mouse SIPA1.
- For ELISA, consider using the antibody as a capture or detection reagent following standard titration to determine optimal working concentrations.
- Lysates from lymphoid tissues (e.g., thymus, spleen) or cell lines with known SIPA1 expression may serve as positive controls.
