Biological Background: Sin3A Function and Localization

  • Encoded by the SIN3A gene, Sin3a (Paired amphipathic helix protein Sin3a, histone deacetylase complex subunit Sin3a) is a transcriptional corepressor that mediates repression through interactions with various transcription factors.
  • Acts as a corepressor for REST, interacts with MXI1 to repress MYC-responsive genes and antagonize MYC oncogenic activities, and interacts with MXD1-MAX heterodimers to tether SIN3A to DNA.
  • Cooperates with OGT to repress transcription in parallel with histone deacetylation; involved in circadian rhythm control by repressing circadian target genes like PER1 through histone deacetylation as part of the large PER complex.
  • Cooperates with FOXK1 to regulate cell cycle progression likely by repressing cell cycle inhibitor gene expression (By similarity).
  • Required for cortical neuron differentiation and callosal axon elongation (By similarity).
  • Located in the nucleus and nucleolus.
  • Highly expressed in the developing brain, particularly in the ventricular zone of various cortical regions.
  • Post-translational modifications include acetylation, phosphorylation, isopeptide bond formation, and ubiquitin-like conjugation (based on UniProt keywords).

Experimental Guidance and Technical Tips

  • The antibody was raised against a synthetic peptide corresponding to the N-terminal region (aa 1-120) of human Sin3A. For applications requiring native conformation (e.g., IP, IF), confirm that the epitope is accessible under native conditions.
  • For Western blotting, the predicted molecular weight is 145 kDa. Use appropriate gel percentage and transfer conditions for high-molecular-weight proteins.
  • If using for immunoprecipitation, consider cross-linking the antibody to beads to avoid interference from the heavy chain which co-migrates near 145 kDa.
  • For immunofluorescence, nuclear localization supports staining patterns; permeabilization may be necessary for nucleolar access.
  • The antibody detects human, mouse, and rat Sin3A, suggesting conserved epitopes; for other species, check sequence homology.
