Biological Background: Sin3A Function and Localization

  • Paired amphipathic helix protein Sin3a (Sin3A) is a ~145 kDa transcriptional corepressor and histone deacetylase complex subunit, encoded by the SIN3A gene.
  • Sin3A functions as a transcriptional repressor, interacting with REST, MXI1, and MXD1-MAX heterodimers to tether the repressor complex to DNA and antagonize MYC oncogenic activities.
  • Sin3A cooperates with OGT to mediate transcriptional repression in parallel with histone deacetylation, and is required for the repression of circadian target genes such as PER1 through the large PER complex.
  • Sin3A localizes to the nucleus and nucleolus, consistent with its role in chromatin remodeling and transcriptional regulation.
  • In the developing brain, Sin3A is expressed at highest levels in the ventricular zone of various cortical regions and is required for cortical neuron differentiation and callosal axon elongation.
  • Sin3A is subject to acetylation, phosphorylation, and ubiquitination, and mutations in SIN3A are associated with disease variants including dwarfism and intellectual disability.
  • The protein contains paired amphipathic helix (PAH) domains, coiled-coil regions, and repeat sequences, facilitating protein-protein interactions within corepressor complexes.
  • Sin3A is involved in the control of circadian rhythms by mediating histone deacetylation-dependent repression of clock-controlled genes.

Experimental Guidance and Technical Tips

  • Consider using a standard SDS-PAGE protocol optimized for high-molecular-weight proteins when performing Western blotting, as the predicted molecular weight of Sin3A is approximately 145 kDa.
  • For ELISA applications, validate the antibody with recombinant Sin3A protein or target-expressing cell lysates and include a negative control.
  • Given the reported cross-reactivity with human and rat, verify specificity in the relevant species by using appropriate positive and negative cell lines or tissues.
