Biological Background: SIK3 Function and Localization
- SIK3 (Serine/threonine-protein kinase SIK3), also known as Salt-inducible kinase 3 (SIK-3) or QSK, is a member of the AMPK-related kinase family and plays key roles in metabolic signaling and cytoskeletal regulation.
- Positive regulator of mTOR signaling that functions by triggering the degradation of DEPTOR, an mTOR inhibitor. Involved in dynamic regulation of mTOR signaling in chondrocyte differentiation during skeletogenesis. Related references: PMID:30232230
- Negatively regulates cAMP signaling pathway possibly by acting on CRTC2/TORC2 and CRTC3/TORC3, thereby influencing glucose metabolism and transcription.
- Prevents HDAC4 translocation to the nucleus, thereby modulating gene expression programs important for cell survival and differentiation.
- Subcellularly localized to the cytoplasm, consistent with its role as a cytoplasmic kinase.
- Expressed in chondrocytes and implicated in skeletal development; mutations in SIK3 have been associated with skeletal dysplasias.
- Contains characteristic kinase domain signatures, including ATP-binding and magnesium coordination sites, and is subject to phosphorylation and methylation.
- Multiple isoforms exist due to alternative splicing, which may contribute to functional diversity.
Experimental Guidance and Technical Tips
- The immunogen encompasses amino acids 60–310 of human SIK3, a region that includes part of the kinase domain; this polyclonal antibody may recognize multiple splice variants—validate with appropriate controls.
- For Western blot, a band around 145 kDa is expected in whole‑cell lysates; consider testing samples from chondrocyte-like cell lines or those with active mTOR signaling.
- Human, mouse, and rat cross‑reactivity is reported, making this antibody suitable for cross‑species comparison studies—optimize conditions for each species.
- ELISA applications may require direct coating of the recombinant immunogen or antigen capture; titer and condition optimization is recommended.
