  • SIK1 (also known as MSK, SNF1LK, or salt-inducible kinase 1) is a serine/threonine-protein kinase that phosphorylates downstream targets including HDAC4, HDAC5, PPME1, SREBF1, and CRTC1/TORC1.
  • It regulates hepatic gluconeogenesis and lipogenesis by phosphorylating and inhibiting the CREB coactivators CRTC1/TORC1 and CRTC2/TORC2, as well as SREBF1, thereby suppressing CREB-mediated transcription in response to cAMP signaling. Related references: PMID:29211348
  • SIK1 is involved in muscle cell differentiation and cardiomyogenesis by phosphorylating class II histone deacetylases HDAC4 and HDAC5, leading to activation of MEF2 target genes and regulation of cell cycle exit.
