Biological Background: SIGLEC6 Function and Localization

• SIGLEC6, also referred to as CD33 antigen-like 1 (CD33L), CDw327, or obesity-binding protein 1 (OB-BP1), is a member of the sialic acid-binding Ig-like lectin (SIGLEC) family.
• It functions as a putative adhesion molecule mediating sialic-acid dependent binding to cells, specifically recognizing α-2,6-linked sialic acid; the binding site may be masked by cis interactions with sialic acids on the same cell surface.
• The protein is predominantly expressed in the placenta (cyto- and syncytiotrophoblastic cells) and at lower levels in the spleen, peripheral blood leukocytes (mainly B-cells), and small intestine.
• Subcellular localization includes the cell membrane and secreted forms, consistent with its adhesion and potential signaling roles.
• The protein contains multiple immunoglobulin-like domains, a transmembrane helix, and undergoes alternative splicing and N-linked glycosylation.

• For western blot, the expected band size is approximately 50 kDa; consider using lysates from human placenta or B-cell lines where SIGLEC6 is endogenously expressed and validate in your specific experimental system.
