Biological Background: SIGLEC10 Function and Localization
- Sialic acid-binding Ig-like lectin 10 (Siglec-10) is a member of the immunoglobulin superfamily that mediates sialic acid-dependent cell adhesion, preferentially recognizing α2,3- or α2,6-linked sialic acids on glycoconjugates.
- Functions as an inhibitory receptor in immune cells, undergoing ligand-induced tyrosine phosphorylation to recruit phosphatases such as PTPN6/SHP-1 via SH2 domains, thereby dampening B-cell antigen receptor signaling and other activation pathways. Related references: PMID:11284738 PMID:12163025
- In association with CD24, modulates natural killer (NK) cell immune responses and may suppress reactivity to danger-associated molecular patterns (DAMPs) like HMGB1, HSP70, and HSP90. Related references: PMID:25450598
- Multiple isoforms exist due to alternative splicing; isoform 5 is the most abundant in peripheral blood leukocytes, while isoform 4 is specifically expressed in NK cells. Found in lymph node, lung, ovary, appendix, bone marrow, spleen, and brain.
