Biological Background: Siglec-8 Function and Localization

  • Siglec-8 (Sialic acid-binding Ig-like lectin 8), also known as SAF2 or SIGLEC8L, is a member of the sialoadhesin family of immunoglobulin-like lectins that mediate cell-cell adhesion through recognition of sialylated glycans.
  • Siglec-8 acts as an adhesion molecule mediating sialic acid-dependent binding to blood cells, preferentially recognizing α-2,3-linked sialic acid with possible cis-masking on the same cell surface. Related references: PMID:10625619 PMID:10856141
  • The protein simultaneously binds terminal sialic acid and underlying 6-O-sulfated galactose residues, showing high affinity for 6-sulfated sialyl-Lewis X glycans, which may direct immune cell interactions. Related references: PMID:27357658
  • It is a type I transmembrane glycoprotein with an extracellular ligand-binding domain, a single-pass transmembrane helix, and a short cytoplasmic tail.
  • Siglec-8 is specifically expressed on the surface of blood eosinophils, basophils, and mast cells, and is subject to alternative splicing.
  • Associated UniProt keywords: 3D-structure, alternative splicing, cell adhesion, disulfide bond, glycoprotein, immunoglobulin domain, lectin, membrane, transmembrane helix, reference proteome.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (aa 17-363) of human Siglec-8, covering the ligand-binding region; this design may enable detection of both membrane-bound and soluble forms.
  • For Western blotting, a band near the predicted molecular weight of 54 kDa is expected; reducing conditions are recommended to resolve monomeric Siglec-8.
  • In ELISA, consider using the antibody to capture or detect recombinant Siglec-8 or native protein from cell lysates; optimization of coating and blocking conditions is advised.
  • Validate specificity by testing on Siglec-8-expressing cells (e.g., eosinophils) versus negative controls, and confirm cross-reactivity with mouse/rat orthologs in the relevant experimental context.
