Biological Background: Siglec-8 Function and Localization
- Siglec-8 (Sialic acid-binding Ig-like lectin 8) is a member of the sialoadhesin family, also known as SAF-2. It is a single-pass type I membrane protein encoded by the SIGLEC8 gene.
- Functions as a putative adhesion molecule that mediates sialic-acid dependent binding to blood cells. Preferentially binds to alpha-2,3-linked sialic acid and, to a lesser extent, alpha-2,6-linked sialic acid. The sialic acid recognition site may be masked by cis interactions with sialic acids on the same cell surface. Related references: PMID:10625619 PMID:10856141
- Recognizes simultaneously epitopes having a terminal N-acetylneuraminic acid (sialic acid) and an underlying 6-O-sulfated galactose, with a preference for Gal-6-sulfated sialyl-Lewis X glycan epitopes. Related references: PMID:27357658
- Expressed specifically on blood cells, namely basophils, mast cells, and eosinophils, making it a key marker for these cell types.
- Subcellularly localized to the plasma membrane.
- Contains immunoglobulin-like domains and is subject to alternative splicing and glycosylation, as indicated by its multiple isoforms and post-translational modifications.
