Biological Background: SHP2 Function and Localization
- SHP2 (Tyrosine-protein phosphatase non-receptor type 11, also known as PTP-1D, PTP-2C, SH-PTP2, SH-PTP3) is a ubiquitously expressed protein tyrosine phosphatase containing two SH2 domains and a catalytic phosphatase domain.
- SHP2 acts downstream of various receptor and cytoplasmic protein tyrosine kinases to participate in signal transduction from the cell surface to the nucleus. Related references: PMID:10655584, PMID:14739280, PMID:18559669
- Positively regulates the MAPK signal transduction pathway and dephosphorylates key signaling intermediates such as GAB1, ARHGAP35, and EGFR. Related references: PMID:28074573

Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein spanning amino acids 514–593 of human SHP2 (C-terminal region), which may recognize multiple SHP2 isoforms. Validate the antibody’s reactivity with the target in your experimental system.
- For Western blotting, titrate the antibody to obtain optimal signal-to-noise ratio using a lysate known to express SHP2 (e.g., HeLa or Jurkat cell lysates).
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimize fixation and antigen retrieval protocols, and include appropriate controls.
