Biological Background: SHMT2 Function and Localization
- SHMT2 encodes mitochondrial serine hydroxymethyltransferase, also known as glycine hydroxymethyltransferase and serine methylase, a key enzyme in one-carbon metabolism.
- It catalyzes the reversible conversion of serine and tetrahydrofolate to glycine and 5,10-methylenetetrahydrofolate, providing one-carbon units for purine biosynthesis and other pathways. Related references: PMID:24075985, PMID:25619277, PMID:29364879
- SHMT2 contributes to de novo mitochondrial thymidylate biosynthesis, crucial for preventing uracil accumulation in mitochondrial DNA. Related references: PMID:21876188

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 265–504 of human SHMT2 (C-terminal region), which may allow detection of full-length and some annotated isoforms. Consider validating reactivity against expected splice variants if necessary.
- For Western blotting (WB), the predicted molecular weight is ~56 kDa, but actual migration may vary depending on gel conditions and post-translational modifications. Include appropriate positive and negative controls.
- Immunohistochemistry (IHC-P) may require antigen retrieval and optimization of antibody dilution. Testing on control tissues with known SHMT2 expression is recommended.
- The antibody is suitable for ELISA; consider using it to quantify SHMT2 levels in cell or tissue lysates following standard sandwich or indirect ELISA protocols.
- Cross-reactivity with mouse and rat SHMT2 is indicated, enabling translational studies. However, individual lot validation in the relevant species is advised.
