Biological Background: Sonic hedgehog (SHH) Function and Localization
- SHH is a member of the hedgehog family, produced as a precursor protein that includes signaling and catalytic domains; the precursor is also known as ShhNC.
- The precursor undergoes autocatalytic cleavage between residues 198-199, releasing the N-terminal signaling domain (ShhN) with a C-terminal cholesterol moiety, while the C-terminal domain (ShhC) catalyzes the reaction and is subsequently degraded in the endoplasmic reticulum. Related references: PMID:7736596, PMID:7891723, PMID:8824192, PMID:21357747
- ShhN acts as a morphogen by binding to the Patched receptor (PTCH1 or PTCH2), which relieves SMO inhibition and activates the smoothened signaling pathway, essential for numerous developmental processes. Related references: PMID:11389830, PMID:31657721, PMID:14687547, PMID:29622726
- During development, SHH signaling controls ventral neural tube and floor plate specification, limb bud anterior-posterior patterning, somite formation, and axon guidance. Related references: PMID:11430830, PMID:12679031, PMID:15315762, PMID:24863049
- The full-length protein localizes to the endoplasmic reticulum and Golgi apparatus membranes; ShhN is secreted and found at the cell membrane, while ShhC remains ER-associated. Related references: PMID:21357747
- SHH is expressed in multiple embryonic tissues such as notochord, ventral neural tube, floor plate, lung bud, zone of polarizing activity, and limb mesenchyme; in adults, it is detected in lung and neural retina.
- The protein contains a signal peptide, zinc- and calcium-binding motifs, and undergoes palmitoylation; it is categorized as a glycoprotein, lipoprotein, and developmental protein.
Experimental Guidance and Technical Tips
- This polyclonal antibody was raised against a recombinant protein spanning amino acids 187-281 of mouse SHH, a region that includes the autoprocessing cleavage site. For Western blot, multiple bands may be observed corresponding to the full-length precursor (approx. 48 kDa) and processed forms such as ShhN (approx. 20 kDa) and ShhC (approx. 25-30 kDa). Validate the antibody in your specific sample and experimental context.
- For ELISA applications, this antibody can be explored as a capture or detection reagent. Since it is a polyclonal preparation, pairing with a monoclonal antibody against a different epitope may enhance assay specificity. Titration and optimization in your assay conditions are recommended.
- The antibody cross-reacts with human, mouse, and rat SHH, making it suitable for studies across these species. However, always confirm specificity via appropriate controls.
