Biological Background: SHC4 Function and Localization
- SHC-transforming protein 4 (SHC4), also known as RaLP or hShcD, is a member of the SHC adaptor protein family. It contains an SH2 domain and a phosphotyrosine-binding domain, mediating interactions with activated receptor tyrosine kinases and downstream effectors.
- SHC4 activates both Ras-dependent and Ras-independent migratory pathways in melanomas, contributing to tumor cell invasion and metastasis.
- It participates in the early phases of agrin-induced tyrosine phosphorylation of CHRNB1 (acetylcholine receptor β1 subunit) at the postsynaptic membrane, linking extracellular matrix signals to synaptic receptor clustering.
- SHC4 localizes to the postsynaptic cell membrane, consistent with its role in synaptic signaling and melanoma migration.
- Tissue expression is highly restricted: SHC4 is only expressed in melanomas, with weak expression in normal melanocytes and benign nevi. Its expression strongly increases at the transition from radial to vertical growth phase and in metastatic melanomas, correlating with acquisition of migratory and invasive competence.
- The protein undergoes alternative splicing and is regulated by phosphorylation, which modulates its adapter functions.
- Molecular weight of SHC4 is approximately 69 kDa.
Experimental Guidance and Technical Tips
- The immunogen, recombinant N-terminal fragment (aa 1-185), may detect multiple SHC4 isoforms resulting from alternative splicing. Consider running a full-range ladder to identify main bands.
- For Western blot, a prominent band near 69 kDa is expected in melanoma cell lines; validate antibody specificity using positive and negative control samples due to restricted expression.
- In immunofluorescence or ICC, anticipate membrane or synaptic localization; co-staining with membrane markers can confirm specificity.
- Reported cross-reactivity with mouse suggests utility in murine models, but homology should be assessed; test in your sample system to verify reactivity.
- ELISA assays may require careful titration of coating antigen and antibody concentration; consider indirect ELISA format for detection.
- For all applications, confirm band specificity and signal-to-noise ratio using appropriate blocking and washing conditions; non-optimal results may necessitate antibody titration or antigen retrieval steps.
