Biological Background: SHBG Function and Localization
- SHBG, also known as sex steroid-binding protein (SBP), testosterone-estradiol-binding globulin (TeBG), and androgen-binding protein (ABP), is encoded by the SHBG gene (GeneID: 6462). Alternatively spliced isoforms 1 and 2 are expressed in liver and testis.
- Functions as an androgen transport protein, specifically binding 5-alpha-dihydrotestosterone, testosterone, and 17-beta-estradiol with high affinity, thereby regulating the plasma metabolic clearance rates of steroid hormones.
- May also participate in receptor-mediated processes, with each dimer binding one molecule of steroid.
- The protein is secreted and features post-translational modifications including glycosylation and disulfide bonds, as well as signal peptide and lipid-binding/steroid-binding repeats.

- IHC-P: Antigen retrieval steps may be required for formalin-fixed paraffin-embedded sections. Evaluate staining in positive control tissues (e.g., liver, testis) and include isotype controls.
