Biological Background: SH2D1A Function and Localization
- SH2 domain-containing protein 1A (SH2D1A), also known as signaling lymphocytic activation molecule (SLAM)-associated protein (SAP), is a cytoplasmic adapter protein critical for immune cell signaling (O60880).
- SH2D1A regulates receptors of the SLAM family, including SLAMF1, CD244, LY9, CD84, SLAMF6, and SLAMF7, acting as a key modulator of lymphocyte activation.
- Early studies proposed that SH2D1A blocks the association of SLAMF1 with inhibitory effectors SHIP1 and SHP-2, preventing negative signaling (PubMed:11806999). Related references: PMID:11806999
- Subsequent work revealed that SH2D1A instead recruits the tyrosine kinase FYN to phosphorylate and activate SLAMF1, initiating downstream signaling (PubMed:12458214). Related references: PMID:12458214
- SH2D1A positively regulates natural killer (NK) cell cytotoxicity by enhancing conjugate formation with target cells through receptors such as CD244, CD84, CD48, SLAMF6, LY9, and SLAMF7.
- It may also modulate signaling of neurotrophin receptors NTRK1, NTRK2, and NTRK3, suggesting roles beyond the immune system.
- SH2D1A is predominantly cytoplasmic and exhibits tissue-specific expression, with high levels in thymus and lung, lower in spleen and liver, and is present in T-lymphocytes; expression is reduced in rheumatoid arthritis.
- Mutations in SH2D1A are linked to X-linked lymphoproliferative disease type 1 (XLP1 or Duncan disease), an often-fatal immunodeficiency characterized by abnormal responses to Epstein-Barr virus infection.
