Biological Background: SGSH Function and Localization
- SGSH encodes N-sulphoglucosamine sulphohydrolase, also known as sulfoglucosamine sulfamidase or sulphamidase, a lysosomal enzyme involved in heparan sulfate catabolism.
- The enzyme catalyzes a key step in the degradation of heparan sulfate by hydrolyzing N-sulfated glucosamine residues, essential for lysosomal clearance of glycosaminoglycans.
- SGSH is localized to the lysosome, where it functions in an acidic environment to break down complex carbohydrates.
- The protein is a glycoprotein with disulfide bonds and binds calcium, which may be important for structural stability or catalytic activity.
- Mutations in SGSH are associated with mucopolysaccharidosis type IIIA (Sanfilippo syndrome A), a severe neurodegenerative lysosomal storage disorder characterized by accumulation of heparan sulfate.
- The precursor protein contains a signal peptide and undergoes post-translational processing to yield the mature enzyme with a calculated molecular weight of approximately 56.7 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 273–502 of human SGSH (NP_000190.1), covering the C-terminal region; thus, the antibody may recognize both precursor and mature forms of the protein, but epitope specificity has not been mapped—validate in your sample system.
- For Western blot, SGSH migrates at ~57 kDa; using appropriate positive controls (e.g., human cell lysates known to express SGSH) and loading controls is recommended to confirm band identity.
- As cross-reactivity with mouse and rat SGSH is reported, the antibody can be considered for orthologous protein detection, but optimal conditions should be determined empirically.
- In ELISA applications, the polyclonal nature may provide broad reactivity and sensitivity; however, include proper negative controls to assess non-specific binding.
