Biological Background: SGOL1 Function and Localization
- Shugoshin 1 (SGOL1), also known as serologically defined breast cancer antigen NY-BR-85 or shugoshin-like 1, is encoded by the SGO1 gene and belongs to the shugoshin family. Multiple isoforms exist due to alternative splicing; isoform 3 has a specialized role in centriole cohesion.
- SGOL1 plays a central role in mitotic chromosome cohesion by preventing premature dissociation of the cohesin complex from centromeres after prophase, ensuring faithful chromosome segregation.
- It acts by protecting the STAG2 subunit of cohesin from phosphorylation at the centromere until cohesin is cleaved by separase at anaphase onset, a process facilitated by its interaction with PPP2R1A.
- Phosphorylated SGOL1 is required for proper chromosome congression and attachment of spindle microtubules to the kinetochore; it is also necessary for the kinetochore localization of PLK1 and CENPF.
- SGOL1 may contribute to the tension-sensing mechanism of the spindle assembly checkpoint by modulating PLK1 kinetochore affinity.
- Subcellularly, SGOL1 localizes to the nucleus, centromere, kinetochore, spindle pole, centrosome, and nuclear speckles.
- It is widely expressed with highest levels in testis, and is strongly overexpressed in approximately 90% of breast cancers.
- Key post-translational modifications include phosphorylation, and the protein contains a coiled-coil domain; it is annotated with keywords such as cell cycle, mitosis, centromere, phosphoprotein, and disease variant.

- For western blotting and immunofluorescence/immunocytochemistry, validate endogenous target expression in proliferating cell lines (e.g., HeLa) where mitotic chromosomes are actively segregating.
- Cross-reactivity with mouse and rat orthologs is predicted; titration and localization patterns should be confirmed in those species.
- In ELISA-based assays, the polyclonal nature may provide robust signal but may require careful blocking and negative controls to minimize background.
