Biological Background: SGMS1 Function and Localization

  • SGMS1 (Sphingomyelin synthase 1) is also known as Medulla oblongata-derived protein, Protein Mob, SMS1, and TMEM23.
  • It is the major sphingomyelin synthase at the Golgi apparatus, catalyzing phosphocholine transfer from phosphatidylcholine to ceramide to produce sphingomyelin and diacylglycerol. Related references: PMID:14685263 PMID:17449912
  • The enzymatic reaction is reversible, with direction governed by the relative concentrations of ceramide and diacylglycerol in Golgi membranes. Related references: PMID:14685263 PMID:14976195 PMID:17449912
  • SGMS1 specifically converts newly synthesized ceramide, delivered from the ER by the CERT transporter, to sphingomyelin. Related references: PMID:30242129
  • It forms a heteromeric complex with glucosylceramide synthase (GCS), which increases SMS activity and reduces glucosylceramide synthesis, controlling the metabolic fate of ceramide. Related references: PMID:30242129
  • Subcellularly, SGMS1 localizes to the Golgi apparatus membrane.
  • SGMS1 regulates signal transduction through the production of signaling lipids (DAG and ceramide) as well as sphingomyelin, a key component of membrane rafts. Related references: PMID:14976195 PMID:17449912 PMID:17982138
  • It also participates in secretory transport by modulating the DAG pool at the Golgi and downstream PRKD1 signaling. Related references: PMID:18370930 PMID:21980337
  • The protein is widely expressed across tissues and undergoes alternative splicing; it is annotated as a phosphoprotein, indicating post-translational regulation.

Experimental Guidance and Technical Tips

  • The immunogen used to produce this antibody covers the N-terminal region (amino acids 1-134) of human SGMS1. For optimal detection, consider using it in the indicated applications: Western blot, immunofluorescence/immunocytochemistry, and ELISA.
  • Predicted cross-reactivity with human and rat SGMS1 is supported by the data; validation in other species is recommended if needed.
  • When performing Western blot, expect a band around 49 kDa; minor variations may occur depending on sample preparation and post-translational modifications.
  • Adjust antibody dilution and incubation conditions based on your specific experimental setup; typical starting dilutions for polyclonal antibodies can be tested.
