Biological Background: SGK3 Function and Localization
- SGK3 (serum/glucocorticoid-regulated kinase 3), also known as cytokine-independent survival kinase (CISK) or SGK-like (SGKL), is a serine/threonine-protein kinase belonging to the SGK family. It plays a pivotal role in regulating ion channels, membrane transporters, and cellular processes such as growth, proliferation, survival, and migration.
- The kinase up-regulates a diverse array of ion channels and transporters, including epithelial sodium channels (SCNN1A/ENAC), potassium channels (KCNA3/KV1.3, KCNQ1, KCNH2/HERG), calcium channels (TRPV5/6), chloride channels (BSND), and amino acid/glutamate transporters (SLC1A5/ASCT2, SLC1A3/EAAT1, SLC1A6/EAAT4, SLC1A7/EAAT5). It also modulates Na+/H+ exchanger SLC9A3/NHE3 and Na+/K+ ATPase activity.
- SGK3 phosphorylates downstream targets including NEDD4L and GSK3B, and regulates transcription through FLII phosphorylation, positively influencing estrogen receptor (ER) transcriptional activity. It also negatively regulates ITCH/AIP4, preventing CXCR4 lysosomal sorting.
- Subcellularly, SGK3 localizes to cytoplasmic vesicles, particularly early and recycling endosomes, consistent with its role in membrane trafficking and transporter regulation.
- Human SGK3 is broadly expressed, with highest levels in pancreas, kidney, liver, heart, and brain, and lower in lung, placenta, and skeletal muscle. Notably, its expression is higher in ER-positive breast tumors compared to ER-negative ones.
- The protein contains ATP-binding and kinase domains, and undergoes phosphorylation, as indicated by its annotation as a phosphoprotein. An alternative splicing isoform has been identified (UniProt keyword).
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fusion protein spanning amino acids 1–150 of human SGK3 (NP_037389.4), targeting the N-terminal region. This region may be suitable for detecting both full-length and potential N-terminal isoforms.
- In Western blotting, a band corresponding to the calculated molecular weight of approximately 57 kDa is expected. Validation across human and mouse cell or tissue lysates is recommended to confirm reactivity.
- For ELISA applications, consider using the antibody as capture or detection reagent; optimal dilutions and conditions should be determined empirically.
