Biological Background: SFTPA1 Function and Localization
- SFTPA1 encodes Pulmonary surfactant-associated protein A1, a member of the collectin family of C-type lectins. It is also known as 35 kDa pulmonary surfactant-associated protein, Collectin-4, and Alveolar proteinosis protein.
- In the presence of calcium, SFTPA1 binds surfactant phospholipids and reduces surface tension at the alveolar air-liquid interface, essential for normal respiration. It enhances expression of MYO18A/SP-R210 on alveolar macrophages (By similarity).
- SFTPA1 plays a critical role in innate immunity by recognizing, binding, and opsonizing pathogens such as Mycobacterium tuberculosis, enhancing their clearance by alveolar macrophages. Related references: PMID:17158455, PMID:21203928, PMID:21123169
- It serves as a receptor for Mycoplasma pneumoniae by binding the CARDS toxin. Downregulation of SFTPA1 reduces toxin binding and associated cellular vacuolization. Related references: PMID:15845487, PMID:25139904
- SFTPA1 is secreted into the extracellular space and localizes to the alveolar surface film and extracellular matrix.
- Post-translational modifications include acetylation, hydroxylation, disulfide bonds, and glycosylation. Alternative splicing generates multiple isoforms.
- Defects in SFTPA1 are associated with pulmonary alveolar proteinosis and interstitial lung disease.

- The immunogen corresponds to a central region (aa 100-248) of human SFTPA1, which is conserved across species, supporting the indicated cross-reactivity with rat.
- For Western blotting, expect a band near 26 kDa. Consider using reducing sample buffer to resolve potential disulfide-linked complexes.
- Given the secreted nature of SFTPA1, appropriate sample types include lung tissue homogenates, bronchoalveolar lavage fluid, or cell culture supernatants from relevant cell lines.
