Biological Background: SFRP1 Function and Localization

  • SFRP1 (Secreted frizzled-related protein 1), also known as SARP-2, is a member of the soluble frizzled-related protein family that modulates Wnt signaling through direct interaction with Wnts.
  • It functions as a negative regulator of the canonical Wnt pathway, decreasing intracellular beta-catenin levels and inhibiting WNT1/WNT4-mediated TCF-dependent transcription.
  • In vascular cells, SFRP1 exhibits antiproliferative effects, delaying the G1 phase and entry into S phase, and can induce an angiogenic response in vivo.
  • During kidney development, SFRP1 inhibits tubule formation and bud growth in metanephroi.
  • The protein is secreted and contains a signal peptide, disulfide bonds, and is glycosylated.
  • SFRP1 is widely expressed, with highest levels in heart and fetal kidney, and is absent from lung, liver, and peripheral blood leukocytes. It is also expressed in testis, ovary, fetal brain, smooth muscle cells, and keratinocytes.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 30–314 of human SFRP1, covering the cysteine-rich domain (CRD) and part of the netrin-like domain; this design may detect full-length SFRP1 and possibly some truncated isoforms.
  • For Western blot, a band at approximately 35 kDa is expected; however, glycosylation may cause a higher apparent molecular weight. Validate the antibody in your specific sample system.
  • For IHC-P, antigen retrieval conditions may need optimization; consider testing on known positive tissues such as heart or fetal kidney.
  • For ELISA, the polyclonal nature may provide sensitivity for detecting soluble SFRP1 in serum or culture supernatants; pairing with a validated capture antibody is recommended.
  • Cross-reactivity with mouse SFRP1 is reported; verify sequence homology between human and mouse SFRP1 to confirm reactivity in your model.
