Biological Background: SF3B1 Function and Localization
- SF3B1 encodes Splicing factor 3B subunit 1, also known as SF3b155, SAP 155, and Prp10, the 155 kDa subunit of the SF3B complex within the U2 snRNP spliceosome. Related references: PMID:12234937, PMID:27720643, PMID:32494006
- It directly participates in early spliceosome assembly and mediates recognition of the intron branch site by promoting selection of the branch-point adenosine, the nucleophile for the first step of splicing. Related references: PMID:32494006, PMID:34822310, PMID:36104565
- As part of the SF3B subcomplex, SF3B1 is required for the formation of the pre-spliceosomal 'A' complex through stable binding of U2 snRNP to the branchpoint sequence. Related references: PMID:12234937, PMID:36104565
- SF3B1, together with SF3A, binds upstream of the branch site in a sequence-independent manner, anchoring U2 snRNP to the pre-mRNA. Related references: PMID:12234937
- The protein also contributes to the assembly of the 'E' complex and functions within the minor spliceosome, which processes U12-type introns. Related references: PMID:10882114, PMID:15146077, PMID:33509932
- SF3B1 localizes to the nucleus and concentrates in nuclear speckles, subnuclear domains enriched in splicing factors.
- Post-translational modifications, including acetylation, citrullination, phosphorylation, and ubiquitination, suggest multiple layers of regulatory control over its splicing activity.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide mapping to amino acids 1–100 of human SF3B1; the antibody may recognize the N-terminal region and is predicted to react with the full-length ~146 kDa protein.
- For western blot applications, consider using a low-percentage SDS-PAGE gel and optimize transfer conditions for high-molecular-weight proteins. Include appropriate molecular weight markers.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), a predominantly nuclear staining pattern with speckled enrichment is expected; validate fixation and antigen retrieval protocols in your tissue or cell system.
- Cross-reactivity with mouse and rat orthologs suggests high sequence conservation in the immunogen region; however, individual experimental verification is recommended.
- ELISA performance should be validated using the recombinant protein or peptide corresponding to the immunogen region as a positive control.
