Biological Background: SF3A3 Function and Localization
-
SF3A3 (Splicing Factor 3A Subunit 3), also known as SF3a60, SAP61, PRP9, or PRPF9, is a 59 kDa component of the SF3A subcomplex within the U2 small nuclear ribonucleoprotein (snRNP) of the spliceosome (UniProt Q12874).
-
It plays a central role in pre-mRNA splicing as part of the 17S U2 SnRNP complex, which directly participates in early spliceosome assembly by recognizing the intron branch site and promoting selection of the branch-site adenosine, the nucleophile for the first transesterification step. -
SF3A3 contributes to the formation of the pre-spliceosome E complex and the pre-catalytic spliceosome A complex, and it remains associated through the formation of pre-catalytic spliceosome B complexes. Related references: PMID:10882114, PMID:11533230, PMID:29360106, PMID:30315277
-
Subcellularly, SF3A3 is localized to nuclear speckles and the nucleoplasm, consistent with its role in splicing factor storage and active transcription sites.
-
The protein is ubiquitously expressed across tissues and is subject to post-translational modifications including phosphorylation and acetylation, as identified by proteomics studies.
-
SF3A3 contains zinc-finger domains and coordinates zinc ions, indicating metal-binding functions that may stabilize protein structure or mediate interactions within the spliceosome.
-
The SF3A complex, composed of SF3A1, SF3A2, and SF3A3, is essential for U2 snRNP recruitment and stable association with pre-mRNA; deficiencies in SF3A3 disrupt splicing fidelity and can lead to aberrant transcripts.
Experimental Guidance and Technical Tips
-
The immunogen is a synthetic peptide mapping to the N-terminal region (aa 1-100) of human SF3A3. For Western blotting, a specific band near 59 kDa is expected; we recommend using nuclear extracts or whole-cell lysates from human (e.g., HeLa, HEK293) or mouse cell lines. Pre-validation of the antibody in the relevant sample system is advised.
-
For ELISA applications, the peptide immunogen can be used as a coating antigen; however, detection of native SF3A3 in complex samples may require optimization. Consider pairing with a detection antibody targeting a non-overlapping epitope.
-
Cross-reactivity has been reported for human and mouse; confirm reactivity in other species if required, as sequence homology varies.
-
Use appropriate negative controls (e.g., siRNA knockdown) to verify specificity in your experimental context.
