Biological Background: SETMAR Function and Localization
- SETMAR is a fusion protein derived from a SET domain histone methyltransferase and the transposase domain of an Hsmar1 transposon, also known as Metnase.
- It catalyzes histone H3 methylation at lysine 4 (H3K4) and lysine 36 (H3K36), and specifically mediates dimethylation of H3K36 at DNA double-strand break sites, which is critical for repair protein recruitment. Related references: PMID:16332963, PMID:21187428, PMID:22231448

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 20-300 of human SETMAR, encompassing the N-terminal region including part of the SET domain. This region may offer good accessibility for detection in IHC-P applications, but formalin fixation could mask epitopes; antigen retrieval may be necessary.
- As a polyclonal antibody, it can recognize multiple epitopes, potentially providing robust signal in ELISA and IHC-P, but batch-to-batch variability should be considered. Titration is recommended for optimal working dilution.
- Suitable for human samples only; cross-reactivity with other species has not been reported. Positive and negative controls should include relevant human tissue or cell lines.
