Biological Background: SETDB1 Function and Localization
- SETDB1, also known as ESET or KMT1E, is a histone-lysine N-methyltransferase that specifically trimethylates lysine 9 of histone H3 (H3K9me3), a hallmark of transcriptional repression. Related references: PMID:11959841, PMID:12869583, PMID:14536086
- It functions primarily in euchromatin to silence genes, coupling H3K9me3 with DNA methylation through interactions with MBD1 and ATF7IP, and this repression is heritably maintained during DNA replication via CAF-1. Related references: PMID:12869583, PMID:14536086, PMID:27237050
- SETDB1 is recruited to chromatin by TRIM28/KAP1, which is directed by KRAB zinc-finger proteins, and is essential for HUSH-mediated heterochromatin formation and gene silencing. Related references: PMID:24623306, PMID:27732843
