Biological Background: SETD2 Function and Localization
- SETD2 is a histone-lysine N-methyltransferase that specifically trimethylates 'Lys-36' of histone H3 (H3K36me3), a mark associated with transcriptional activation. Related references: PMID:16118227, PMID:19141475, PMID:21526191
- It is the primary H3K36me3 methyltransferase, coordinating FACT complex recruitment and interacting with hyperphosphorylated RNA polymerase II to modulate chromatin during elongation. Related references: PMID:23325844
- SETD2-mediated H3K36me3 is critical for DNA mismatch repair in G1/early S phase by recruiting the MutSα complex to chromatin. Related references: PMID:23622243
- Required for DNA double-strand break repair via homologous recombination, promoting RAD51 recruitment at damage sites. Related references: PMID:24843002
- Acts as a tumor suppressor and is frequently mutated in various cancers. Related references: PMID:24509477
- Also methylates non-histone substrates, including α-tubulin (Lys-40) and STAT1 (Lys-525), influencing mitosis, cytokinesis, and antiviral interferon responses. Related references: PMID:27518565, PMID:28753426
- Ubiquitously expressed and localizes to the nucleus and chromosomes.
- Contains a SET domain and multiple functional motifs; functions as a chromatin regulator and transcriptional activator.
Experimental Guidance and Technical Tips
- For Western blot, use a high-molecular-weight marker and extended transfer times to ensure efficient detection of the 288 kDa SETD2 protein.
- Include appropriate positive and negative controls (e.g., HeLa cell lysates, SETD2 knockdown samples) to confirm band specificity.
- When performing ELISA, consider titrating the antibody against the immunogen or full-length protein to determine optimal coating and detection conditions.
- Given the polyclonal nature, each new lot should be validated in the relevant human sample system to confirm consistent reactivity and specificity.
