- Serpin B6 (also known as CAP, PI-6, PTI, SPI3, etc.) is a member of the serine protease inhibitor (serpin) superfamily, which regulates proteolytic cascades through suicidal inhibition mechanisms.
- It functions as a cytoplasmic inhibitor of serine proteases, including cathepsin G, kallikrein-8, and thrombin, thereby modulating extracellular matrix remodeling, inflammation, and coagulation.
- SERPINB6 is implicated in the protection of the inner ear; loss of its inhibitory activity may lead to leakage of lysosomal contents during stress, resulting in hair cell death and sensorineural hearing loss (DFNB91).
- The protein is primarily localized in the cytoplasm and is widely expressed, with highest levels in skeletal muscle, and also found in placenta, heart, lung, liver, kidney, pancreas, and inner ear hair cells.
- It is expressed in keratinocytes and mast cells, suggesting roles in epithelial barrier function and immune cell proteostasis.
