Biological Background: SERPINB6 Function and Localization

• Serpin B6 (also known as CAP, PI-6, PTI, SPI3, etc.) is a member of the serine protease inhibitor (serpin) superfamily, which regulates proteolytic cascades through suicidal inhibition mechanisms.
• It functions as a cytoplasmic inhibitor of serine proteases, including cathepsin G, kallikrein-8, and thrombin, thereby modulating extracellular matrix remodeling, inflammation, and coagulation.
• SERPINB6 is implicated in the protection of the inner ear; loss of its inhibitory activity may lead to leakage of lysosomal contents during stress, resulting in hair cell death and sensorineural hearing loss (DFNB91).
