Biological Background: Serotonin Transporter Function and Localization

The serotonin transporter (SERT), encoded by the Slc6a4 gene (also known as Htt or Sert), is a sodium-dependent transporter belonging to the solute carrier family 6, member 4. It acts as an antiporter, cotransporting serotonin with Na⁺ in exchange for K⁺, thus mediating the reuptake of serotonin from the synaptic cleft and limiting its signaling. In the central nervous system, SERT is essential for serotonin homeostasis. During development, it controls serotonin uptake in glutamatergic neurons of the somatosensory cortex, guiding proper spatial organization of cortical neurons and sensory circuit formation. - The immunogen corresponds to the N-terminal region (amino acids 1-85) of mouse SERT. For Western blotting, a band at ~70 kDa is expected. Use reducing conditions and appropriate tissue lysates (e.g., brain or lung).
- For immunohistochemistry (paraffin-embedded sections), validate the antibody in known positive tissues (brain or gut) and optimize antigen retrieval conditions.
