Biological Background: SERCA1/ATP2A1 Function and Localization
- Also known as Sarcoplasmic/endoplasmic reticulum calcium ATPase 1, Calcium pump 1, Calcium-transporting ATPase sarcoplasmic reticulum type, fast twitch skeletal muscle isoform, and Endoplasmic reticulum class 1/2 Ca(2+) ATPase.
- Functions as a key regulator of striated muscle performance by acting as the major Ca2+ ATPase responsible for the reuptake of cytosolic Ca2+ into the sarcoplasmic reticulum.
- Contributes to calcium sequestration involved in muscular excitation/contraction. Related references: PMID:10914677
- Localizes to the endoplasmic reticulum and sarcoplasmic reticulum membranes.
- Predominantly expressed in skeletal muscle, specifically in fast twitch (type II) fibers.
- Catalyzes ATP hydrolysis coupled with calcium translocation; binding sites for ATP, calcium, and magnesium are present.
- Undergoes alternative splicing, yielding isoforms with calculated molecular weights around 110 kDa (full length) and 95 kDa.
- The protein is post-translationally modified by phosphorylation and contains disulfide bonds.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a C-terminal region (aa 900-1001); this design may confer isoform specificity. Consider cross-reactivity with other SERCA family members when interpreting data.
- For Western blot, expect bands at approximately 95 kDa and 110 kDa, reflecting alternative splicing isoforms. Optimize gel electrophoresis and transfer conditions accordingly.
- In immunofluorescence protocols, permeabilization is required due to the membrane localization of SERCA1. Validate the staining pattern in relevant positive controls.
- The antibody is validated for human, mouse, and rat samples. Performance in additional species should be verified empirically.
- As a rabbit monoclonal antibody, this reagent may offer high affinity and consistent lot-to-lot reproducibility. Confirm optimal working dilutions for your specific assay and sample type.
