Biological Background: Septin-8 Function and Localization

• Septin-8 is a filament-forming cytoskeletal GTPase that belongs to the septin family of proteins. It plays a role in platelet secretion by participating in SNARE complex formation in synaptic vesicles.

• The protein stabilizes BACE1 protein levels and promotes the sorting and accumulation of BACE1 to recycling or endosomal compartments, thereby modulating the beta-amyloidogenic processing of APP.
• Multiple isoforms exist due to alternative splicing. Isoform 2 is expressed at low levels in specific brain areas, while isoforms 1 and 3 are highly expressed in the occipital pole, frontal lobe, temporal lobe, and putamen. Isoform 2 is highly expressed in prostate, testis, and ovary; isoforms 1 and 3 are expressed at low levels in these tissues.
• Septin-8 localizes to the cytoplasm, cytoskeleton, synaptic vesicles, and presynaptic terminals. It is also found at the synaptic vesicle membrane, axons, and cytoplasmic vesicles.
• Key post-translational modifications include acetylation and phosphorylation, which may regulate its function. The protein contains a GTP-binding domain and a coiled-coil region.
• Widely expressed in tissues, with highest abundance in the aorta, as well as in brain, heart, and platelets.

• The immunogen corresponds to the C-terminal region (amino acids 348-427) of human Septin-8 (NP_001287728.1), which may recognize multiple isoforms. For WB, consider using whole cell lysates from human, mouse, or rat tissues, and validate the expected band at approximately 56 kDa.
• For IHC-P, optimal antigen retrieval conditions should be determined; brain or aorta sections are suitable positive controls given the high expression in these tissues.
• In IF/ICC, anticipate cytoskeletal and punctuate cytoplasmic staining patterns; co-staining with synaptic or vesicular markers can help confirm localization.
