Biological Background: Septin 2 Function and Localization
- Septin-2, also known as NEDD-5, is encoded by the SEPTIN2 gene. It belongs to the septin family of GTP-binding proteins, and alternative splicing generates multiple isoforms.
- It is a filament-forming cytoskeletal GTPase that assembles into higher-order structures, playing critical roles in cytoskeletal organization and cell division.
- During mitosis, Septin-2 forms a scaffold at the midplane of the mitotic spindle, which is required for maintaining CENPE localization at kinetochores and enabling chromosome congression and segregation.

- Septin-2 is required for normal organization of the actin cytoskeleton and contributes to the biogenesis of polarized columnar epithelium by maintaining polyglutamylated microtubules.

- For Western blotting, a band of approximately 41 kDa is expected. Consider including cytoskeleton-enriched fractions in your lysate to enhance detection, as septins are often associated with the cytoskeleton.
