Biological Background: Semaphorin-4D Function and Localization
- Semaphorin-4D (also known as CD100, A8, BB18, GR3) is a single-pass type I membrane protein that exists as both membrane-bound and secreted forms. It belongs to the semaphorin family and contains an immunoglobulin domain.
- Acts as a cell surface receptor for plexins PLXNB1 and PLXNB2, mediating cell-cell signaling important for neurogenesis and immune regulation. Related references: PMID:20877282
- In the nervous system, it modulates neurite outgrowth and arborization via RHOA activation through PLXNB1, and promotes the development of inhibitory GABAergic synapses. Related references: PMID:19788569
- Drives cerebellar granule cell migration and endothelial cell migration via SRC-PTK2B-AKT signaling. Related references: PMID:16055703
- In the immune system, it induces B-cell aggregation and enhances their viability in vitro, highlighting a role in lymphocyte biology. Related references: PMID:8876214
- Highly expressed in skeletal muscle, peripheral blood lymphocytes, spleen, and thymus, with lower expression in brain, kidney, and other tissues; not detected in colon and liver.
- Post-translationally modified by glycosylation and phosphorylation; subject to alternative splicing. Subcellularly localizes to the cell membrane and is also secreted.
Experimental Guidance and Technical Tips
- The immunogen covers the entire extracellular domain (aa 22-734), suggesting the antibody likely detects extracellular epitopes. For flow cytometry, consider using live, non-permeabilized cells to verify surface staining.
- For immunofluorescence/immunocytochemistry (IF/ICC), membrane localization requires proper permeabilization with detergents like Triton X-100 at low concentrations. Validate staining patterns in known Semaphorin-4D-expressing cell lines.
- In ELISA, the antibody may be used as a capture or detection reagent; cross-reactivity with other semaphorin family members should be assessed if working with complex biological samples.
- Due to alternative splicing and glycosylation, the observed protein weight on western blot may vary; if using this antibody for western blot, validate with appropriate controls and consider that the immunogen is recombinant protein.
