Biological Background: Semaphorin-7A Function and Localization
- Encodes Semaphorin-7A, a GPI-anchored membrane glycoprotein belonging to the semaphorin family; also known as CDw108 and JMH blood group antigen.
- Functions in integrin-mediated signaling, promoting focal adhesion complex formation and activation of PTK2/FAK1, leading to MAPK1/MAPK3 phosphorylation.
- Regulates immune responses by inducing pro-inflammatory cytokine production in monocytes and macrophages, and modulating T-cell-mediated inflammation.
- Supports neuronal development by promoting axon growth in the embryonic olfactory bulb.
- Facilitates melanocyte attachment, spreading, and dendrite outgrowth.
- Subcellular localization: anchored to the cell membrane via a GPI moiety; expressed in skin keratinocytes, endothelial cells, and widely in tissues such as placenta, testis, ovary, spleen, brain, spinal cord, lung, heart, adrenal gland, lymph nodes, thymus, intestine, and kidney.
- Contains an immunoglobulin domain and sema domain; subject to alternative splicing and disulfide bond formation.
Experimental Guidance and Technical Tips
- For WB, the expected band is approximately 75 kDa; consider using appropriate membrane blocking and antibody dilution validated in your sample system.
- The immunogen spans the extracellular sema domain (aa 45-280); thus, the antibody may detect both full-length GPI-anchored and shed soluble SEMA7A forms.
- Since SEMA7A is membrane-associated, optimize protein extraction with membrane fraction enrichment protocols for enhanced detection.
- For ELISA, validate the antibody with recombinant SEMA7A protein or appropriate native samples to establish optimal working conditions.
