Biological Background: SEC61B Function and Localization
- SEC61B encodes the beta subunit of the SEC61 translocon complex, a central component of the endoplasmic reticulum (ER) protein translocation machinery. Related references: PMID:12475939
- The SEC61 complex forms a ribosome receptor and a gated pore in the ER membrane, enabling cotranslational import of signal peptide-containing precursor polypeptides. Related references: PMID:12475939
- It also mediates ER membrane insertion of transmembrane proteins, working in concert with the multi-pass translocon (MPT) complex for multi-spanning membrane proteins. Related references: PMID:32820719, PMID:36261522
- SEC61B cooperates with the translocating protein TRAM1 to facilitate nascent protein import into the ER. Related references: PMID:19121997
- Subcellularly, SEC61B is an integral component of the endoplasmic reticulum membrane.
- The protein undergoes several post-translational modifications, including acetylation, palmitoylation, and phosphorylation, which may regulate its activity or stability.
- As a key player in protein translocation, SEC61B is associated with processes such as protein transport, translocation, and ER membrane organization.
Experimental Guidance and Technical Tips
- For Western Blot (WB), the predicted molecular weight of SEC61B is approximately 10 kDa. Use a 12-15% SDS-PAGE gel and optimize transfer to a PVDF membrane with 0.2 µm pore size for small proteins. Include appropriate positive controls (e.g., HeLa, HEK293, or tissue lysates expressing SEC61B) and validate antibody specificity with a blocking peptide or knockout sample.
- For Immunofluorescence/Immunocytochemistry (IF/ICC), fixation with 4% PFA followed by permeabilization with 0.1% Triton X-100 is recommended. Test antibody concentration in the range of 1:50–1:200 and confirm localization at the ER. Co-staining with an ER marker (e.g., calnexin or PDI) can help verify specificity.
- For ELISA, the antibody can be evaluated as a capture or detection reagent. Titrate the antibody to determine the optimal working concentration, and validate with a recombinant SEC61B protein or cell lysate. Ensure cross-reactivity testing with mouse and rat samples to confirm performance.
- As a polyclonal antibody, lot-to-lot consistency should be monitored. Consider aliquoting upon receipt and performing a pilot study for each new lot to maintain experimental reproducibility.
