Biological Background: SEC24C Function and Localization
- SEC24C (SEC24-related protein C) is a component of the COPII coat complex, which drives vesicle formation from the endoplasmic reticulum (ER) for cargo transport to the Golgi apparatus. Related references: PMID:10214955
- It plays a key role in cargo selection, with a specificity profile distinct from SEC24A and SEC24B; together with SEC24D, it may preferentially package GPI-anchored proteins through the cargo receptor TMED10. Related references: PMID:17499046, PMID:18843296, PMID:20427317
- SEC24C specifically recognizes IxM motif-containing cargos, such as the SNARE proteins GOSR2 and STX5, facilitating their incorporation into COPII vesicles. Related references: PMID:18843296
- The protein localizes to the cytoplasmic side of the ER membrane, COPII-coated vesicle membranes, and the cytosol, consistent with its role in vesicle budding.

Experimental Guidance and Technical Tips

- In immunofluorescence/ICC, optimize fixation and permeabilization conditions to visualize ER and COPII vesicle patterns; co-staining with organelle markers is recommended.
- For ELISA, the immunogen (C-terminal fragment) can be used as a positive control; assess performance with recombinant SEC24C or endogenous protein from cell extracts.
