Biological Background: Sec23A Function and Localization
- Sec23A, also known as SEC23-related protein A, is a core component of the coat protein complex II (COPII) that drives transport vesicle formation at the endoplasmic reticulum (ER).
- It participates in both the physical deformation of the ER membrane into vesicles and the selection of cargo molecules for delivery to the Golgi complex.
- Plays a role in insulin signaling by facilitating the translocation of the glucose transporter GLUT4 (SLC2A4) to the cell surface (by similarity).
- Localizes to the ER membrane, COPII-coated vesicle membranes, and the cytosol, reflecting its dynamic role in vesicular trafficking.
- Expressed ubiquitously in human tissues.
- Undergoes post-translational modifications such as acetylation and phosphorylation; contains zinc-finger motifs that may mediate protein–protein or protein–membrane interactions.
- Multiple isoforms are generated through alternative splicing.
- Mutations in SEC23A have been linked to disease (exact variant not detailed in current record).
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-150) of human Sec23A; for western blot, a band of approximately 86 kDa is expected.
- For ELISA applications, plate coating with the full-length recombinant protein or cell lysates from relevant species may be suitable; assay conditions should be optimized empirically.
- Cross-reactivity with mouse and rat is stated; however, users should verify reactivity in their specific sample matrix, e.g., using positive and negative controls.
- As a polyclonal antibody, batch-to-batch consistency should be assessed for quantitative assays.
