Biological Background: SEC13 Function and Localization
- Protein SEC13 homolog is a component of the nuclear pore complex (NPC) and the COPII coat, crucial for nucleocytoplasmic transport and ER-to-Golgi vesicle trafficking. Related references: PMID:8972206
- At the endoplasmic reticulum, SEC13 participates in the biogenesis of COPII-coated vesicles and is required for the efficient export of secretory proteins such as adipsin from the ER.
- As a core member of the GATOR2 complex, SEC13 positively regulates the mTORC1 signaling pathway by inhibiting the GATOR1 subcomplex. Related references: PMID:23723238, PMID:25457612, PMID:27487210, PMID:35831510
- In amino acid-rich conditions, the SEC13-containing GATOR2 complex likely catalyzes ubiquitination of NPRL2, leading to GATOR1 inactivation and mTORC1 activation. Related references: PMID:36528027
- SEC13 and its paralog SEH1L are required for the structural stability of the GATOR2 complex, ensuring optimal mTORC1 modulation. Related references: PMID:35831510
- SEC13 localizes to multiple cellular compartments including the cytoplasmic side of COPII vesicles, endoplasmic reticulum membrane, nuclear pore complex, and lysosome membrane.
- Post-translational modifications include acetylation and phosphorylation, and the protein undergoes alternative splicing.
- SEC13 contains multiple WD40 repeat domains, facilitating protein-protein interactions in multi-subunit complexes such as the NPC and GATOR2.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the full-length human SEC13 protein (residues 1–322). The antibody is expected to recognize all known isoforms generated by alternative splicing.
- For Western blotting, the predicted molecular weight is ~36 kDa. Use appropriate positive and negative controls to validate specificity in your sample system.
- In immunohistochemistry and immunofluorescence/ICC, permeabilization steps may be critical due to the membrane-associated and nuclear localizations of SEC13. Optimize fixation and permeabilization conditions.
- The antibody shows cross-reactivity with human, mouse, and rat samples; however, we recommend verifying reactivity in your specific model system.
- As a polyclonal reagent, lot-to-lot consistency should be monitored. Consider performing a bridging study when introducing a new batch into an established assay.
