Biological Background: SDSL Function and Localization
- SDSL (Serine dehydratase-like), also known as cSDH, SDH 2, or TDH, is a pyridoxal-phosphate-dependent enzyme that catalyzes the dehydrative deamination of L-serine and L-threonine to pyruvate and alpha-ketobutyrate, respectively. Related references: PMID:16580895 PMID:18342636
- It also exhibits glutamate racemase activity, interconverting L-glutamate and D-glutamate (By similarity).
- SDSL is predominantly expressed in lung cancer cell lines, linking it to tumor metabolism.
- The enzyme functions as a lyase and isomerase, and is involved in amino acid and lipid metabolism.
- It is subject to acetylation and contains a pyridoxal phosphate binding site.
- The molecular weight of the canonical isoform is approximately 35 kDa (theoretical: 34.7 kDa).
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fusion protein spanning amino acids 1–137 of human SDSL. This N-terminal region may include key catalytic residues; consider that the antibody may recognize epitopes within this domain.
- For Western blotting (WB), use appropriate cell or tissue lysates expressing SDSL, such as lung cancer cell lines, and validate the detected band at the expected molecular weight (~35 kDa).
- In ELISA applications, plate coating with the recombinant immunogen or peptide fragments can serve as positive controls.
- Given reported cross-reactivity with mouse, the antibody may be suitable for orthologous studies, but always verify in the relevant mouse sample system.
- Pyridoxal phosphate-dependent enzymes can be sensitive to reducing agents; optimize loading buffer conditions as needed.
