Biological Background: SDPR/Cavin-2 Function and Localization

  • SDPR (Serum deprivation-response protein), also known as caveolae-associated protein 2 (cavin-2), PS-p68, and phosphatidylserine-binding protein, is encoded by the CAVIN2 gene in humans.
  • Cavin-2 plays an important role in caveolar biogenesis and morphology by inducing membrane curvature within caveolae. Related references: PMID:19525939
  • It is required for caveola formation in a tissue-specific manner, being essential in lung and fat endothelia but not in heart endothelia (by similarity).
  • Cavin-2 negatively regulates the size or stability of CAVIN complexes in lung endothelial cells.
  • The protein may play a role in targeting protein kinase C alpha (PRKCA) to caveolae (by similarity).
  • Subcellular localization includes cytoplasm, cytosol, membrane, and specifically caveolae, a type of lipid raft.
  • Post-translational modifications include acetylation and phosphorylation, as indicated by proteomics and direct protein sequencing evidence.
  • Expression is high in heart and lung, with lower levels detected in brain, kidney, liver, pancreas, placenta, and skeletal muscle.

  • The immunogen is a synthetic peptide from the C-terminal region (amino acids 326-425); consider using tissues with high expression (heart, lung) or positive cell lines as controls.
  • For Western Blot, the predicted molecular weight is approximately 47 kDa. Use standard denaturing and reducing conditions; confirm band specificity with knockout or knockdown samples if available.
  • In immunofluorescence and immunohistochemistry, cavin-2 localizes to the plasma membrane and cytosol. Permeabilization may be necessary for intracellular detection.
  • For ELISA, titrate the antibody with recombinant protein or peptide to establish a linear detection range.
  • Reported cross-reactivity includes human and mouse; verify reactivity in other species as sequence homology outside the immunogen region may vary.
