Biological Background: SDHD Function and Localization
- SDHD is a membrane-anchoring subunit of succinate dehydrogenase (complex II) in the mitochondrial inner membrane, essential for complex stability and function. Related references: PMID:10482792, PMID:9533030
- It plays a critical role in the electron transport chain by transferring electrons from succinate to ubiquinone (coenzyme Q). Related references: PMID:10482792, PMID:9533030
- SDHD also contributes to malate oxidation, converting malate to enol-oxaloacetate, a potent inhibitor of succinate dehydrogenase, which is then isomerized into keto-oxaloacetate. (By similarity)
- The protein is localized exclusively to the mitochondrial inner membrane, consistent with its predicted transmembrane domains.
- SDHD is known by multiple aliases, including PGL1, CII-4, and QPs3; mutations in the SDHD gene are linked to hereditary paraganglioma syndromes. (Keywords: Disease variant)
- As part of complex II, SDHD connects the tricarboxylic acid cycle to the mitochondrial respiratory chain, underscoring its metabolic importance. (Keywords: Tricarboxylic acid cycle)
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 50–150 of human SDHD. Peptide competition assays can be used to verify antibody specificity.
- For Western blotting, SDHD migrates at ~17 kDa; using a 12–15% polyacrylamide gel and appropriate molecular weight standards is recommended.
- This antibody has been validated for WB and ELISA; optimal working dilutions should be determined empirically for each application and sample type.
- Cross-reactivity with mouse and rat samples has been demonstrated, but validation in the specific experimental model is advised.
