Biological Background: SDHB Function and Localization
- SDHB encodes the iron-sulfur (IP) subunit of succinate dehydrogenase (SDH), also known as mitochondrial respiratory chain complex II, which transfers electrons from succinate to ubiquinone (PubMed:26925370, PubMed:27604842). Related references: PMID:26925370 PMID:27604842
- SDH additionally catalyzes the oxidation of malate to enol-oxaloacetate, a potent SDH inhibitor, which is subsequently isomerized into keto-oxaloacetate.
- The protein is nuclear-encoded, synthesized with a transit peptide, and imported into the mitochondrial inner membrane, where it integrates into complex II.
- SDHB contains [2Fe-2S], [4Fe-4S], and [3Fe-4S] iron-sulfur clusters essential for electron transfer and belongs to the oxidoreductase family.
- Aliases such as PGL4 and MC2DN4 reflect associations with paraganglioma and mitochondrial complex II deficiency, underscoring its role in disease.
- Post-translational modifications include acetylation, and the protein is a component of the tricarboxylic acid (TCA) cycle and electron transport chain.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 29–280 of human SDHB, encompassing the majority of the mature protein. Under reducing SDS-PAGE conditions, a specific band at approximately 32 kDa is expected.
- For IHC-P, antigen retrieval (e.g., heat-mediated citrate buffer) and titration of the primary antibody on positive control tissues (e.g., skeletal muscle, heart) are recommended.
- In ELISA, optimize coating antigen concentration and antibody dilution to achieve the desired sensitivity and specificity.
- For Western blot, consider using mitochondrial-enriched lysates or high-polarity tissues to enhance signal detection.
- Validate antibody performance in your experimental system, as cross-reactivity with other species or subcellular components may vary.
