Biological Background: SDHB Function and Localization
- SDHB encodes the iron-sulfur (Ip) subunit of mitochondrial succinate dehydrogenase (Complex II), which is a key enzyme linking the Krebs cycle and oxidative phosphorylation.
- Core function: transfers electrons from succinate to ubiquinone (coenzyme Q) in the electron transport chain (PubMed:26925370, PubMed:27604842). Related references: PMID:26925370 PMID:27604842
- Also catalyzes the oxidation of malate to enol-oxaloacetate, a potent SDH inhibitor that isomerizes to keto-oxaloacetate (By similarity).
- Alternative names include Iron-sulfur subunit of complex II, SDH, SDH1; the gene is also known as SDH, CWS2, PGL4, among others.
- Subcellular localization: integral component of the mitochondrial inner membrane.
- The protein contains multiple iron-sulfur clusters (2Fe-2S, 3Fe-4S, 4Fe-4S) essential for its electron transfer function and is subject to acetylation.
- Associated with primary mitochondrial disease; mutations in SDHB are linked to hereditary paraganglioma and pheochromocytoma (Keywords: Disease variant, Primary mitochondrial disease).
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 29–280 of human SDHB, corresponding to the mature protein after mitochondrial import and processing; thus, the antibody is suitable for detecting endogenous SDHB across human, mouse, and rat samples.
- For Western blotting, an expected band at ~32 kDa can be observed. Use appropriate mitochondrial extraction or lysis buffers (e.g., RIPA with protease inhibitors) to ensure efficient solubilization of this integral membrane protein.
- In immunofluorescence/ICC applications, consider permeabilization with Triton X-100 or digitonin and validate mitochondrial co-localization with markers such as MitoTracker or anti‑COX IV.
- For ELISA, the antibody can be used for capture or detection of SDHB in biological fluids or cell lysates; optimize coating conditions and antibody concentrations for your specific assay.
- As with all polyclonal antibodies, lot-to-lot variability may occur; we recommend titrating the antibody in your system and including appropriate controls.
