Biological Background: SDHB Function and Localization
- SDHB encodes the iron-sulfur (Ip) subunit of succinate dehydrogenase (SDH), also known as mitochondrial respiratory chain complex II.
- The SDH complex catalyzes the oxidation of succinate to fumarate in the tricarboxylic acid (TCA) cycle and transfers electrons to ubiquinone via its iron-sulfur clusters (PubMed:26925370, PubMed:27604842). Related references: PMID:26925370 PMID:27604842
- The protein coordinates [2Fe-2S], [3Fe-4S], and [4Fe-4S] clusters that mediate electron transport.
- SDHB is anchored in the mitochondrion inner membrane with its active site facing the matrix.
- In addition to succinate, SDH can oxidize malate to enol-oxaloacetate, which acts as a potent inhibitor of the complex (By similarity).
- Acetylation and other post-translational modifications regulate SDH activity, and mutations in SDHB are linked to primary mitochondrial disease and hereditary paraganglioma/pheochromocytoma syndromes (based on UniProt keywords: Disease variant, Primary mitochondrial disease).
- The protein is also noted for host-virus interactions and is a target of proteomic identification.
Experimental Guidance and Technical Tips
- The immunogen spans the N-terminal region (aa 29–280) of human SDHB, which includes the mature protein after mitochondrial processing. Verify antibody specificity in mitochondrial fractions or purified complex II.
- For Western blotting, SDHB migrates at ~31 kDa; use reducing conditions and consider adding protease inhibitors to prevent degradation.
- In immunofluorescence or IHC, permeabilization is required to access the mitochondrial inner membrane; validate staining with a mitochondrial marker.
- The antibody cross-reacts with mouse and rat SDHB; confirm sequence homology if using other species.
- For ELISA, pair with a validated capture/detection system targeting a non-overlapping epitope if developing a sandwich assay.
